Summary

Human cytochrome P450 3A4 and CYP3A5 are major P450 enzymes in the liver and gastrointestinal tract. They play important roles in the metabolism of a wide variety of drugs and some endogenous steroids. In this study, we investigated the behavior of CYP3A activity and the protein expression of CYP3A4 and CYP3A5 in volunteers who were repeatedly administered a St. John’s wort caplet (300mg) three times a day for 3 days. CYP3A activity in the volunteers was measured by urine and mRNA samples, and protein concentration of CYP3A4 and CYP3A5. The results showed that CYP3A activity exhibits different behavior among humans. The expression content of CYP3A4 and CYP3A5 associated with CYP3A activity also differs among humans.
CYP3A activity cannot be explained by SNP of the CYP3A enzyme. CYP3A activity was different depending on the adjustment of CYP3A4 and CYP3A5 expression.
